A SNF2-like protein faciliates dynamic control of DNA methylation

چکیده
Supplementary Methods Primers used for genotyping: The α’ promoter silencing system and the drd1 mutant alleles have been described previously (Kanno et al., 2004). The work reported here was carried out using plants carrying the drd1-6 allele. The drd1-6 mutation produces a G to A substitution (TGGTCA to TGATCA), producing a new NdeII restriction enzyme site (/GATC) that can be used for genotyping after PCR amplification with the following primer set: 5’ AGC TAA GGG ATG GAA ACT AGG 3’ 5’ CGA GAT GCT CCA ACA AGC GCG 3’

A SNF2-like protein facilitates dynamic control of DNA methylation.
DRD1 is a SNF2-like protein previously identified in a screen for mutants defective in RNA-directed DNA methylation of a seed promoter in Arabidopsis. Although the initial study established a role for DRD1 in RNA-directed DNA methylation, it did not address whether DRD1 is needed for de novo or maintenance methylation, or whether it is required for methylation of other target sequences. We show...

In plants, the mechanism by which RNA can induce de novo cytosine methylation of homologous DNA is poorly understood. Cytosines in all sequence contexts become modified in response to RNA signals. Recent work has implicated the de novo DNA methyltransferases (DMTases), DRM1 and DRM2, in establishing RNA-directed methylation of the constitutive nopaline synthase promoter, as well as the DMTase M...

Background: Endometriosis (which affects up to 50% of infertile women) is one of the major causes impacting female infertility. Endometriosis, defined as the presence of endometrial glands and stroma outside the uterine tissue, causes a wide range of functional disorders in the process of follicular development and changes in the follicular milieu, resulting in the formation of an incompetent o...
متن کاملO-11: N-a-acetyltransferase 10 Protein Regulates DNA Methylation and Embryonic Development
Background Genomic imprinting is a heritable and developmentally essential phenomenon by which gene expression occurs in an allele-specific manner1. While the imprinted alleles are primarily silenced by DNA methylation, it remains largely unknown how methylation is targeted to imprinting control region (ICR), also called differentially methylated region (DMR), and maintained. Here we show that ...
متن کاملSNF2 chromatin remodeler-family proteins FRG1 and -2 are required for RNA-directed DNA methylation.
DNA methylation in Arabidopsis thaliana is maintained by at least four different enzymes: DNA methyltransferase1 (MET1), chromomethylase3 (CMT3), domains rearranged methyltransferase2 (DRM2), and chromomethylase2 (CMT2). However, DNA methylation is established exclusively by the enzyme DRM2, which acts in the RNA-directed DNA methylation (RdDM) pathway. Some RdDM components belong to gene famil...
